We sell shirts and someone placed an order for 5 of them. So, one order of five shirts.

Order with shirts is wrong then?

It's fine. Or "an order." Or send "them" out today, if you are thinking of the 5 items.

    I am not a teacher.

The order can also be 'for' or 'of' five items.

Can I say " I got a business today. The customer order five of items."?

No, Polyester. Both your sentences are wrong.

Batman45, if you're the seller, I would say "Today, we got an order for five items".
Remember - if you don't use correct capitalisation, punctuation and spacing, anything you write will be incorrect.
