1- By 2050, there will have been an additional two billion, most of them from poor countries.
2- By 2050, there will be an additional two billion, most of them from poor countries.

I have no problem with the first one but I would like to ask if the second one is OK or not.

It's the other way round for me. I have a problem with the first.

#2 is just fine.

It depends entirely on what the original sentences refer to. If it refers to deaths from malnutrition, then 1. is correct. The same for births, funerals and marriages.
I agree with Tarheel that, if it refers to people, 'people' should be included, and 2 is correct.
