    Is the following sentence grammatically correct?

    "Kelly, I am James, from the Leadership Course."

    Leadership course​ is not a proper noun, so it should not be capitalized. With that corrected, the sentence will be grammatically correct.
    I am not a teacher.

    If the name of the course is "Leadership Course", it would be capitalised (in a list of courses, at least). However, I agree that it doesn't necessarily need to be capitalised in the OP's sentence. Presumably both the speaker and the listener know which leadership course the speaker is referring to.
    Remember - if you don't use correct capitalisation, punctuation and spacing, anything you write will be incorrect.

    In writing, I would omit the second comma ... unless James pauses and has to add the last bit when Kelly shows no sign of recognition.

    I agree with Rover about the second comma, though I don't consider it to be wrong.
