This IS the best experience OF/IN my life.
Which preposition sounds more natural in BE? Or are both correct from a grammar point of you? I'd prefer "of". Thanks a lot in advance!

Your bigger problem is the missing verb in that sentence.

The verb is not missing: This IS...: present simple of to be!

I'd use "of". However, I'm more likely to actually say "This is the best experience I've ever had".

I would say "this is the best thing I've ever experienced."
Otherwise, use "in".

I think of is a better choice, as it tends to be preferred in phrases like this where we have a short event followed by a longer, periodic event which contains it. We understand the "experience" as a moment belonging to the larger period of a "life[time]". Similarly, we use phrases like

the hottest day of the year
the low-point of my career
the highlight of the match
I had the time of my life
