What does reach mean here? I checked the dictionary and guessed it might be

the limit within which someone can achieve something.

But why is it funny?

Your guess is right.

DiCaprio is not noted for being full of self-doubt in real life. He was asked if it was very hard to prepare to play the part of an insecure actor.

It's not very funny. They laughed mainly out of politeness.

After some stumbling, the interviewer asks Was that a reach? He effectively means Was that difficult for you?

They laugh because Leo Di Caprio is an actor full of self-doubt (as they all are), so it obviously was not difficult for him to imagine playing one. The question is sarcastic.
