    What is the difference between a lot of and lots of?

    I think the latter is colloquial and in this register denotes a larger number than the former.

    I have a lot of grammar books.
    I have lots of grammar books - more than just a lot.

    It seems in formal speech lots of is inappropriate.

    Still I think we want a native speaker's help, Winston.

    As a native speaker, I don't notice any difference between the two.

    He's got a lot of money.
    He's got lots of money.

    To me they seem to be two different ways of saying the same thing. But you may be right that "lots of" is inappropriate in formal speech (or writing).
