    I take it that you are asking about these as terms for quantity:

    A number of cows have escaped from the field = some cows (unquantified number) have escaped from the field

    The number of cows that have escaped is 12 = 12 cows have escaped

    Any amount of cows can escape = there is no saying how many cows may or may not escape; it could be one and it could be 100.

    Also:

    The amount of money in his account is substantial = he has a lot of money in his account
