    Which one is correct?

    1- What day is it today?
    2- What day is today?

    Must we necessarily use "it" in this context?

    You generally would use 1. if you were asking whether it was Monday or Tuesday, etc. (The stress is on "day"). 2 is simply not colloquial for this.
    You could 2 if the answer was "my birthday", ie. if you were asking "What is special about today". The stress would be on "today" - but even then, 1 is more common for this too.
