    what's the meaning of this sentence?

    He left home to seek his fortune in St.Louis.

    "He left home to seek his fortune in St.Louis."
    He left for St. Louis looking to make a lot of money.

    Examples of seek and fortune:

    I visit UsingEnglish.com to seek answers to my questions.
    So "seek", in context, means "to look for", "to look for answers".

    I made a fortune selling peanuts, now I can afford a nice car.
    "Fortune", in context, means "a lot of money".

    Yes it can. That's how I'd read it. But it could mean either.

    Yes, it can mean a lot things.
    I don't know the original source of the saying "to seek one's fortune". It obviously means different things to different people. "Destiny" sounds the best so far.
