I have some queries? Can anybody help me?

-"He's no friend of mine".Why is "no " used here?
-What does this sentence mean:"It takes a nation of millions to hold us back".What structure is used?

It's a construction that emphasises that he's not your friend. It means he is definitely not your friend in any way at all, and possibly an enemy.
You could think of it as: "He is no [kind of] friend of mine".
    Consider this conversation:
    A: I thought C was a doctor.
    B: C is not a doctor.
    (a straightforward assertion that C isn't a doctor)
    B: C is no doctor.
(He is certainly not a doctor! Nor is he a dentist, nor a vet. In fact he has had no medical training at all. He's a complete fake.)
