I wanted to call Dr Johnson to book an appointment; however I dialled the incorrect telephone number and called Dr. Mary's office by mistake.

I'd use the comma. Be consistent with your punctuation. Otherwise it's OK.

(1) Here in the United States, most people feel that a comma is mandatory after "however."

(2) Therefore, you might want to write:

book an appointment; however, I dialed the incorrect....
