Don't = contraction of "do not"
Doesn't = contraction of "does not"

Doesn't would be used with the 3rd person singular, while don't would be used with all other forms.

I don't know.
He doesn't have any coffee.
We don't go to the cinema very often.
Don't you know the answer?
Doesn't she have to go to school every day?
Don't do that!
It doesn't take very long to walk from my house to the park.
