I'm not sure if this verb is properly used: Every one of you are unique.
or shall I use "Every one of you is"?

"One" is singular. One is.

"Every one of you is unique." Don't let the "you" throw you off. "Of you" is a prepositional phrase. "One" is the subject of the sentence.
