In English we say " how do you do? " when meeting someone for the first time.. right!
But
Do tour guides have to say it when introducing themselves to the tourists they meet?
or they can just say;" Hi, my name is bla.. bla.. bla.."?

No - they are probably better to start: "Good morning, my name is xxxx and I am your guide for xxxx".

To some extent, this can depend on the nature of the tourist group.
If the group is mostly young people or teenagers they would probably warm to you more if you use "Hi........", but if the group is older, the more formal greeting suggested by Anglika would be preferable.
