Hello,
What does the word keeper mean in this sentence?
From all guys she has dated this one is the keeper.
(That's not how exactly they said it, but I cannot find the scene anymore.)

Is keeper in this case a bad or a good word?
I tried to use some dicts but I don't see the sense.

It means that she is interested in continuing her relationship with him.

Not a teacher.

Think about fishing. You catch a fish -- it's too small. You throw it back. You catch the next fish -- it's big enough. You keep it. That second fish is "a keeper."

Now think of it as a metaphor for dating. It's not that they are too small or large enough, but good enough so you don't want to throw that one back and try to hook another.

The first one with the right structure of an interogative sentence, is correct .

Either one. You have the verb following the subject in both so the position of "in this sentence" can be flexible.
