I see this word often while reading novels. But I find difficult to use it in my conversation. how it is different from "inspite of" or "but" . Please let me know its usage properly.

You don't need to use it in speech. I'd use 'still, anyway, anyhow'.
Still, you could use 'nevertheless' if you wanted to.
Nevertheless, it would sound a bit formal.
Anyway, there are simpler words.
