It depends upon the context whether "Do you want to play with me?" is sexual or not.

How about you giving us some examples of the contexts in which you have met it?

hahaha
You know!! I thinks I have a bit similar situation. I was calling all boys who I talk with: My boyfriend. An I thought that boyfriend means a lover and a normal friend. When my friend says to me who is he?, I just say: he is my boyfriend. After some days and when she asks me about other guy: who is he?, I say he is my boyfriend as well :D. How shameful. But now I knew that the word boyfriend means a lover.
Also, I was calling my frinds(girls) my girlfriend, so people laughed at me a lot. And a lot of them thought that I'm a ***.:-( What a pity.
Anyway I call my lover my sacred man. Or God's angel.

Possible contexts:

"do you want to play with me?"
An invitation to engage in a game of some sort (tennis, golf, cards, etc.) or perhaps a musical performance. Have not heard it used in a sexual sense.

"do you want to have sex with me?"
One meaning and one meaning only.

"do you want to get off with me?"
Get off (the bus, subway, etc.). More likely, a reference to "getting high" (as in drug use), which could lead to something sexual. Have not heard it otherwise used in a sexual sense (that would be more like "Get your rocks off").

"do you want to tap off with me?"
Totally new. Never before encountered.
