'Vanity plates' are car licence plates where the owner can choose the word they want, like their name, or similar. I'm not entirely sure that it is a particularly funny cartoon.;-)

I saw the cartoon and didn't think it was funny. At any rate, if something (such as success) goes to your head, it makes you feel more important or better than you really are.
