I was watching an old crime-thriller film last night.

One of the gangsters suddently enters a room with a gun in his hand. He points the gun at the men that were there playing cards (a group of 5 cops) and says just: "Reach!" Reach!"

Is it the same in this context as "Don't move!"? or is it "similar to "Hands up!"? or maybe it's slang for something else like "Stand up!"

The abovementioned movie is American and it was produced in the forties.

It means 'Hands up!'

In those days, gangsters (such as Hugh Doidy-Ratte) used to shout 'Reach for the sky!' or 'Reach!' for short.
