I am wondering if my sentences are grammatically correct.

The serious illness of his little daughter affected John deeply. From the moment he woke up until he went to sleep his only thought was how to cure her.

The construction of the first half, "The xxx of his xxx", is unnatural.

John's little/young daughter's serious illness affected him deeply.

You might wonder why, in that case, I started with "The construction of the first half". The difference is that your example involved people so the usual possessive construction (with an apostrophe and an s) is more natural.
