"What's up?"

It means how's everything, am I right?
Can I say it to my boss?

Thank you.

It is very informal.

People's relationships with their boss wouldn't usually allow this degree of informality.

Note my corrections above. As Roman55 indicated, it entirely depends on your relationship with your boss. I would happily use it with my boss but I work in a very informal atmosphere where, with the exception of our individual responsibilities, we treat each other as equals.

It can be used both ways in AmE, either to inquire about a problem, or ask about what is happening in general.

Well, I would guess you don't speak English to your boss in Asia.

I think there are some Asian countries where English is spoken in workplaces to some extent, such as Singapore, India, Malaysia and Philippines,
