    For a start, it should be "It is I..."

    If it's I who am right.(in this case, without a doubt, is right)
    But the thing I want to know( if we use the informal english) is if we can say : It's me(accusative) who am right.( instead of me who is right).

    There is not much agreement on this, as you'll see here:
    WHO
    Subject-verb agreement when "who" is involved
