Please is it / də ʌp / or / du ʌp /?

If these are transcriptions of do up, then only the second is correct. In BrE, we normally transcribe it as /du: ʌp/

:up: (/ə/ is always unstressed. If you stress a vowel, it can't be /ə/ .
