As for Steve, he crashes a bomber jet into the arctic to keep Hydra from destroying New York. He is also presumed dead, but he survives, frozen in suspended animation for the next 66 years. Jeez, doesn't anyone who dies in this movie actually die?

Hello, teachers. The speaker apparently makes a point that the characters in this movie, who are supposed to die, survive. So I would expect a positive question:

Does anyone who dies in this movie actually die?

What's the point in a negative question using "Doesn't"?

The "doesn't" is more emphatic and incredulous.

"Does anyone here know how to make a good cup of coffee?" is a neutral question.

"Doesn't anyone here know how to make a good cup of coffee?" expresses something different.
