I am a host of an English club meeting. Before the start of our last meeting, I asked participants a question:

How many of us have already been vaccinated?

Is the above italic sentence natural? And how about the following italic sentence, is it natural too?

How many of you have already been vaccinated?

It depends if you want to make a point of including yourself. If you only want to know about the other people in the room, use "you". If you want to know about everyone in the room, including yourself, use "us".

You could go with "Put your hand up if you've been vaccinated". That way, you can put your hand up or not, as appropriate. Bear in mind that some people are not comfortable disclosing their vaccination status. Is there a reason you want to know?

A case of cultural difference here? The reason is that it's very common for people here to ask if one's been vaccinated.

The government encourages people to get vaccinated due to the new cases from time to time. So in my online groups we can see organizers asking people to get vaccinated.

I asked because I want to know where and how to get vaccinated. Due to the recent outbreak in Guangdong, we needed to wait two hours to get vaccinated but there are some places where we don't need to wait.

Yes. It's not as inclusive as "us," of course, but it's still natural.

I like the "us" version better.

They're good, but they're not better. They seem a bit abrupt for a conversation starter. I still like your first version best.

Taken literally, yes, it does. But a native speaker would not take it literally. I would be understood as Silver's way of including himself in the discussion.
