Hello!

Could someone please explain if there's a significant difference between 'a stone fireplace in the center of the room' or 'a stone fireplace at the center of the room'?

Absent any context, there's no difference in meaning to me. Using "in" is more natural and common in BrE. However, as is always the case, the rest of the sentence, the surrounding sentences and the context might lead to a different answer.
