'Which of you is the best English speaker?'

Other alternatives are possible.

Just use the ones Rover gave.
 
However,

"Who can speak English more fluently, you or Ken?"

I wouldn't use 'which' in the construction above.

Is 'which' acceptable there? (No, I don't think so.)
