My question : Can we construct a sentence as witten below?
She didn't love me and rely on me.
or
She didn't love me and not rely on me.
Thanks...

She didn't love me and rely on me. No.

She didn't love or rely on me.
She neither loved nor relied on me.
She didn't love me, nor did she rely on me.

Re: conjunction, and
She didn't love or not rely on me.
Can we put 'not' in here?

No, because then you have a double negative (and we all know about those!) It would read "She didn't not rely on me" and that's not grammatically correct.

Re: conjunction, and
Ok, I think I have understood.
We can't put 'not' at both sides of a sentence with a conjunction such as (and, or,..etc). Is that ok? Is one 'not' enough for one side?
She didn't love or didn't rely on me.

She didn't love or didn't rely on me.
You don't need "didn't"" twice either, unless you make it two almost separate sentences as you did in your very first example:
She didn't love me, and she didn't rely on me.

*NOT A TEACHER*
I think we can you as I will write now ,but I am not sure whether I am right or not.,
He ate the chocolate ,but didn't like it.
We can use like that because of the fact that 'not' is only at the right side of the sentence. Is that correct? Or is it related to 'but' ?

Remove the comma and your sentence is fine. The only omission is the word "he" before "didn't" - that omission is absolutely fine.
Review your punctuation/spacing. The comma goes directly after a word, then you put a space, not the other way round. Don't put a space before a question mark. Always put a space after a full stop.

Re: conjunction, and
It would be 'use' not 'you'. :)
Must we write 'didn't' or is it enough if we write only 'not' instead of 'didn't ' at this sentence?
He ate the chocolate but not like it.

You need "did not" or "didn't" or you don't have a complete verb in the negative form.
