I watched a movie with only one actor in it.

Can we put emphasis "Least" by adding "Most" or "More"?

Since least and most are opposites, it would be difficult.

No. "Least" is a superlative so you can't intensify it with "more", and "most" is the opposite of "least".

Is it correct to say "fewest number of actors" do I need to strictly stick to "fewest actors" and "largest actors"? Later doesn't sound fine. Am I correct?

Oops! Sorry. I didn't mean you should replace the word least with the word fewest. I meant we'd phrase it differently. Using "least number of" isn't natural.

Using "smallest number of" would be better. But it would be more natural to say something like:

- What movie you've seen had the fewest actors?
- Of all the movies you've seen, which one had the fewest actors?

"The fewest actors" is fine. "The largest actors" are the ones who are physically the biggest. John Goodman is a large actor.

A number can't be fewer than some other number. Just say "What movie have you seen with the fewest actors?" Mine is My Dinner with Andre, with only two.

Perhaps:

What movie that you have watched had the most actors?

(Who cares?)
 
Probably a war film- the fewest may well have some artistic interest.
