Is sentence number 2 also correct? How about in the past tense? What would be the correct verb to use?

1. It is I who am thankful.
2. It is I who are thankful.

"I am".
("I are" is an incorrect conjugation since "are" always refers to a plural noun or pronoun.)

The verb "to be" would be the correct one.
