e.g)where would you get off telling him that you are proud of him?
2. Look* who’s talking?

Please edit your post and put the relevant words from your title somewhere in your post. You haven't used "Who's talking?" in any context in the post. "What's the difference between them" shouldn't appear in your title at all. A good title would have been "Where would you get off telling him that you are proud of him?" That would ensure that your title is unique.
 
The difference between 1 and 2 is that they have absolutely no discernible connection with each other.
Thank you! I thought both meant “you don’t have a right to talk to person A like that” (like a student talking to their own teacher in a teacher-like manner)
 
"Where do you get off talking to ... like that?" is nothing like "Look who's talking!"

The first means "How dare you talk to ... like that?" The speaker has potentially been rude or disrespectful.

The second means something along the lines of "You're being a hypocrite!" It means that the other speaker has said something about someone else that could just as easily be applied to the speaker. For example:

John (who is wearing very ugly shoes): Oh my god! Look at that bloke's shoes! They're the ugliest things I've ever seen.
Helen: Look who's talking!
