Which one is correct?

-Do you know who is the richest person in the world?

-Do you know who the richest person in the world is?

Sentence 2 is correct. You only use the construction "Who is/are....." when you are asking a direct question, and "who" is the first word of the sentence.

Who are you?
Do they know who you are?
I know who you are?

Who was the first man on the moon?
Do you know who the first man on the moon was?
I know who the first man on the moon was.

Note that this is true of the verb "to be".

With other verbs:

Who broke the vase?
Do you know who broke the vase?
I know who broke the vase.

Who ate all my cookies?
Do you know who ate all my cookies?
I know who ate all my cookies.
