use of word "busybee"

Do we say "busybee" when we only refer to a girl,eg:How are you busybee (because she is always busy)
What word do we use when we want to refer to a guy as busy.

No, a bee is either male or female.

You've sure been a busy bee! Look at all you've gotten accomplished!

That could be said to either a male or female, but it's quite informal.
 
Because bee is feminine opposite of bee is Drone

According to a rather sweet kids' website about bees, these are the following names:

Collective noun - bee
Male bee - drone
Female bee - queen, worker
Baby bee - Larva
Group name - Hive, swarm (only when in flight), bike, drift, grist.

Busybee applies to both sexes!
