You must take at least a 1-hour lunch break

I would say:
You must at least take a one-hour lunch break.

Not a teacher

I would say that none of these is correct. While the third one seems natural enough, 'at least' says to me that the lunch break must last a minimum of one hour. What you would probably hear is 'You must take a one hour lunch break' or '...take one hour for lunch'.

I could accept #3 (if you had numbered them) but 'at least' is probably not what is meant. You wouldn't want the employee to have the lunch break stretch to two hours or three. You might as well give him every afternoon off.
