In which case, can we completely do away with Ram’s help on this matter and filing be done at Kolkata office with your help?

What does "In which case" mean here? Can we use "In this case" instead?
 
No. "Which" is asking for information. "This" is not.
 
If it is asking for information, should the comma after 'In which case' be omitted?
 
Can't we begin the sentence with "Can"? How does it make any difference to begin with "in which case". Please clarify
 
'In which case' refers back to what was written earlier, so we need to see at least the previous sentence.
