Sir,I want to know, Is this a correct sentence?

I want to know if this is a correct sentence.
It is not correct.

Perhaps you mean 'I will be taking up/resuming my duties on 16 April, 2016'.

Without more context, it's hard to say.

Dear Sir, Please correct the following sentence-

"I will be joining office on 16 April 2016."

I would use the/your office, or say something like I will be starting work at your office on 16 April.

I have merged the two threads. As you can see, ivarsharma, you had already been told in the first thread not to address us all as "Sir" yet you went on to do it again in the second thread.

Please make sure you follow the advice you are given in this forum.
 
Without more context, it's hard to say.

But it's not too hard to say that join your duties is unlikely to be correct in any context- you can join meetings, etc, but not duties IMO.
