People who have no experience at/with show business often underestimate the power of media. Should I say at or with?

Don't use "at". If you say "experience with", it means that the people have at some time come into contact with show business. I suggest you say "experience in" or "experience of" - that suggests that those people have actually worked in show business at some time.
