They are completely different!

A supermarket's main purpose is selling food although some bigger supermarkets sell other things (clothes, electronics).

A department store is a large shop which has many different sections. Most department stores sell clothes from lots of different companies/designers etc. They might also have a large section selling household items, another selling kitchenware, another selling lighting. Some department stores also have a food section.
 
Along with what emsr2d2 has said, there are "super" centers in the US that combine both a supermarket and a discount department store.

But, in general, department stores may sell some food, but not fresh meat or produce.
