I wonder if this sentence should be changed to: don
t let your tear be the last drop of water on the earth.

No, it shouldn't. 'Drop' should be changed to 'drops'.

But you've given no reason why you want to change it.
 
It depends on who is crying. You could use drops instead of drop, but it is not a massive distinction- one teardrop or five, it's still the last water on earth.
