"When answering the question "what is the most unusual place that you have heard of?"
Can I start my sentence with present perfect and say "I have heard about/of the glowing lake in the Maldives. It glows due to the life of the activity of microscopic organisms."

Yes, the present perfect would be the most natural tense, given that it's used in the question. Bear in mind that in natural conversation, a native speaker might not bother with that opening at all.

John: What's the most unusual place you've ever heard of?
Sarah: An amazing glowing lake in the Maldives!
