Can I use "in a difficult moment" instead of "in a difficult time" or " in a difficult situation"?

The best friend is someone who will help you in a difficult moment.

At a difficult time (not 'in') = when you are facing a difficult situation.

In difficult times = During a period when you're going through difficulties in general.

Yes, you can use these two.
