Here are some examples of sentences.

I am a mechanic in a garage.
He is a waiter in a restaurant.
She is a teacher in a school.

If a specific school were used, would that change the sentence?

Which is correct?

1. He is a lecturer at Harvard University

or

2. He is a lecturer in Harvard University

So in this example sentence number 1 would be correct?

3. He is a lecturer in a university

4. He is a lecturer at a university.

Again, which one is correct?

Both 3 and 4 are correct. I would use 4.
