could anyone please let me know which sentence is correct and why:


1) My both eyes are twitching.

2) Both my eyes are twitching.

if any sentence is not correct please let me know the reason.

"both of my eyes" or "both my eyes"?

Hmm. Probably "Both of" but I have to say that "Both" sound perfectly natural to me. Could be regional variation.
 
You have two different current topics labelled "language". It's best if you call your posts something specific, like "Both my eyes" for this one. This helps with searching for posts later, and with identification of follow-up posts and threads that you are following.

The determiners like, all, both and half, usually precede other attributive adjectives.( The possessive adjectives (my, your, her) are like attributive adjective.)
e.g.
all three tables (not three all tables)
both my legs/eyes/ears. (not my both legs)
half the red roses (not the red half roses)
Here are a few examples:
I feel pain in both my eyes
Left side of both my eyes are red
The difference in the powers of both the eyes is a cause for concern
