Yes, I do.

I understand these are the natural responses to those questions. However, I am just wondering if we can say "Positive" as an answer. It may not be natural but is it grammatical correct?
 
No. It makes no sense.

If you want to sound like a robot, use affirmative for yes and negative for no.
 
Or a cop/soldier.
 
Your second question should be "Do you know how to drive a car?"

If you want a question that has "positive" as a response, try "What type of charge is on a Hydrogen ion?" :lol:
 
Also: 'Are you sure you can be here in time?'

'Positive.'
