I want to know if do could be used to put emphasis on verb do.

Also, please tell me if repetition of "do and did" ONLY makes sense of emphatic sentences or other helping verbs can be repeated to show emphasis.

Eg. I am am a good man. I do am a good man. I have have a pen. I do have a pen. etc.
 
Only 'I do have a pen' is correct.

Yes, it can:

'Mum says I don't do enough jobs around the house, bit I do do lots of chores.'

We don't repeat other helping verbs for emphasis.
 
Just to clarify, it's not a verb repetition. It's just the use of the helping verb "do" (which can be used with most verbs) with the main verb "do".

In spoken English, the helping verb "do" is stressed.

"I do have a pen."
"I do go to that school."
"I do do my homework every night."

The auxiliary "do/did" isn't used with any form of the verb "to be".

I am a man.
Are you a man?

I was a doctor.
Were you a doctor?

I had a pen.
Did you have a pen?

I like cats.
Do you like cats?
 
I used to be a doctor.

Did you use to be a doctor?

Fair point, though I would say the verb there is made up the modal "used to" plus "be". But I can't deny that it fits my "any form of the verb to be".
