I am not a teacher but neither one sounds right to me. Maybe it is because it is unclear who is requiring it.

Correct sentences would include:

We require you to know it.
We are required to know it.
We require the knowledge of it.

'Require' is normally used only in the passive when followed by a to-infinitive:
We are required to know it = it is a requirement (laid down by somebody) that we know it.

If that is not what you mean, you could simply say: we need to know it.
