1. Let's start with exercise N1.
2. Let's start from exercise N1.

I would normally expect "with" in that use.
 
I would normally expect to see 'Let's start with exercise #1' (read as '...exercise number one').

I would say 'let's get started with exercise #1.':)

That works with a capital "L" in "Let's".
