Could you tell me if “ That’s a lot of work.“ and “ That’s lots of work.” are interchangeable? They’re confusing. Please help me. Thank you.
 
They're not interchangeable, You're unlikely to hear native English-speakers say “That’s lots of work.”

Northeast Central U.S. but I'm sure it would be understood in any area of the U.S. without a strange look or double take.

That is is singular and lots of is plural. That doesn't seem right to me, but if the native speakers here say it's OK, then it is OK.
