Can I use this instead of that (in that is my boss) without the change of meaning?

Please correct the timestamp. "14:2" doesn't exist. Do you mean "14:20" or "14:02"?
 
It's true that "this" and "that" are often interchangeable, but here you should stick to "that" for two reasons.
1- The character in the striped t-shirt (B) is actually answering a question asked by the character in the blue suit (A).
A: Who is that?
B: That is my boss.
2- The "boss" is actually not that close in that particular scene, so "that" is more appropriate.
 
This is my boss.

This is appropriate if you're personally introducing your boss and he/she can hear what you're saying.
