I would like to ask you about the preposition that we use with IVR. Should it be "on" or "in"?

As written, you're suggesting that John would say all three of those sentences one after the other. That would be ridiculous. Did you mean to present them as three alternatives and ask us which one would be best?

IVR stands for interactive voice response. It's an automated way of directing a telephone enquiry. Instead of: press 1 for this or 2 for that, an IVR asks questions in English and can understand simple responses like yes and no.
 
To answer your original question, I would use "on," but "in" is possible, too.

Which of those three responses would be best for John to say?
